Melatonin , also known chemically as N-acetyl-5-methoxytryptamine, is a naturally occurring hormone found in most animals, including humans, and some other living organisms, including algae. Circulating levels vary in a daily cycle, and melatonin is important in the regulation of the circadian rhythms of several biological functions. Many biological effects of melatonin are produced through activation of melatonin receptors, while others are due to its role as a pervasive and powerful antioxidant with a particular role in the protection of nuclear and mitochondrial DNA.

Melatonin has been studied for the treatment of cancer, immune disorders, cardiovascular diseases, depression, seasonal affective disorder (SAD), circadian rhythm sleep disorders and sexual dysfunction. Studies by Alfred J. Lewy at Oregon Health & Science University and other researchers have found that it may ameliorate circadian misalignment and SAD. Basic research indicates that melatonin may play a significant role in modulating the effects of drugs of abuse such as cocaine.

Treatment of circadian rhythm disorders
Exogenous melatonin taken in the evening is, together with light therapy upon awakening, the standard treatment for delayed sleep phase syndrome and non-24-hour sleep-wake syndrome. It appears to have some use against other circadian rhythm sleep disorders as well, such as jet lag and the problems of people who work rotating or night shifts.

Taken 30 to 90 minutes before bedtime, melatonin is put into the blood earlier than the brain’s own production and acts as a mild hypnotic.
A very small dose taken several hours before bedtime in accordance with the phase response curve for melatonin in humans (PRC) doesn’t cause sleepiness but, acting as a chronobiotic, does advance the phase slightly and is additive to the effect of using light therapy upon awakening. Light therapy may advance the phase about one to two-and-a-half hours and a small oral dose melatonin, timed correctly some hours before bedtime, can add about 30 minutes to the advance achieved with light therapy.

Procto-glyvenol provides rapid relief from unpleasant symptoms connected with haemorrhoids (pain, tingling, pricking, or tightness of skin). Does not affect the central nervous system or heart function.

Procto-Glyvenol: Tribenoside, a synthetic compound belonging to the group of glucofuranosides, and lidocaine, local anaesthetic (contained in both forms of Procto-Glyvenol), bring a rapid relief of unpleasant symptoms connected with haemorrhoids (pain, tingling, pricking, feeling of tight skin) that are usually caused by a secondary inflammation of the anorectal area. A healing effect of Procto-Glyvenol in patients with haemorrhoids is guaranteed due to its specific activity – first, owing to its ability to decrease permeability of walls of the smallest vessels and to increase the vessel wall tension – and second, owing to its local anti-inflammatory effect and owing to its action against various substances which originate in the inflammatory tissue and which take a joint effect in development of inflammation and pain. The lidocaine component helps to relieve pain quickly. As Procto-Glyvenol is aimed to reach a local effect, it is desirable for the absorption of the active substance tribenoside into the body to be as lowest as possible. Approximately 2-20% of tribenoside will be absorbed by the skin from the cream, from one suppository it is 20-27% of the dose. After absorption tribenoside is subjected to an intensive metabolism in the body and the substances created in this way are excreted into urine. Lidokaine is absorbed in a very small amount and does affect neither the central nervous system, nor the heart function. – DocSimon
